二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

本人专注于肌肉骨骼系统分化及相关疾病的机理研究。以通讯作者身份发现小G蛋白Rac1活性的异常高表达在骨关节炎中的致病机理(Ann Rheum Dis2013)和上游的关键调控机制(AR2015),分子马达介导的胞内转运在肌肉分化中的时空调控作用(MBoC2015),以及热激蛋白对组蛋白基体和细胞增殖的调控功能(JBC2015)。所有研究工作全部在国内完成,并发表在国际主流学会期刊,相关研究成果Nature Reviews Rheumatology 2014. 10:1 作为研究亮点推介。领导的课题组主持肌肉骨骼系统研究领域的国家级课题五项(本人主持四项)。

申请人回国以来,在肌肉骨骼系统的分化及疾病机理研究方面建立了较为完整的研究体系,立足国内发表SCI文章7篇,总影响因子61.3。其中以通讯作者在国际主流学会期刊发表4篇,阐明了骨关节炎的发病机理和肌肉分化再生的分子机制。申请人近年来的主要学术成绩、创新点及科学意义如下:

1.      发现小G蛋白Rac1在骨关节炎中的异常激活,并提供了可能的干预手段。骨关节炎影响人群广,但相关机理不明且缺乏有效的干预手段。申请人和合作者领导的团队发现骨关节中小G蛋白Rac1的异常上调,而且干预Rac1的活性会影响骨关节炎的发展,并利用生物材料缓释Rac1抑制剂在动物体内对骨关节炎起到治疗效果。这一研究为阐明骨关节炎的发病机制做出了重要贡献,也为开发相关干预手段提供了新靶点。工作于2013年在Annals of the Rheumatic DiseasesIF12.38在线发表2015年正式出版)。此外,这项工作在次年被Nature Reviews Rheumatology 2014. 10:1 作为研究亮点(Research Highlights)推介

2.      阐明小G蛋白Rac1在骨关节炎中被异常激活的分子机理。在前一项研究的基础上,申请人和合作者领导的团队深入研究骨关节炎中Rac1被异常激活的分子机理。利用筛选发现骨关节炎中Rac1的负面调控分子OCRL1表达下调,并明确了OCRL1通过影响Rac1的活性对骨关节炎进行调控。该项工作进一步明确了骨关节炎的发表机理,为治疗提供了新思路。该研究成果于2015年被美国风湿病学会期刊Arthritis & RheumatologyIF6.01)接受发表。

3.      明确分子马达蛋白介导的胞内运输在成肌细胞分化中的作用。成肌细胞作为肌肉前体细胞,是肌肉分化发育的基础。申请人发现分子马达蛋白KIF5B在成肌细胞分化过程中上调,并通过动态转运p38MAPK/MKK信号蛋白复合物来调控成肌细胞分化。该项工作揭示了对信号通路进行精准的时空调控在肌肉分化中的重要意义。该研究于2015年被美国细胞生物学会期刊Molecular Biology of the Cell接受发表。

4.      揭示小分子量热激蛋白在细胞增殖中的作用及机理。小分子热激蛋白Cpn10对肌肉发育起着重要作用,但相关的分子机理不明。申请人和合作者发现Cpn10通过和NPAT分子的相互作用来调控细胞核内的组蛋白基体,从而影响组蛋白基因转录和细胞增殖。该研究为肌肉生成提供了潜在的新干预靶点,研究成果于2015年被美国生化学会期刊Journal of Biological Chemistry接受发表。


三、岗位工作思路&= #21450;预期目标

申请人竞聘成功后的工作思路主要包括以下几方面:

1. 教学方面:继续承担医学本科生和研究生核心课程教学任务,以及承担和协调海宁联合学院生物医学课程,将本人主讲的课程做成高质量的精品课程。继续做好海外高水平教学团队的原味课程引进学习工作(以色列希伯来大学发育生物学教学团队等),为培养高素质的医学生和生物医学人才做出贡献;

2. 科研方面:在任职后的未来5年内,按计划完成在研的四项国家级项目基础上,继续深入拓展目前的肌肉骨骼研究方向。目前已发现数个肌腱及肌肉特异性敲基因小鼠在肌肉肌腱分化和代谢方面表现出独特的表型,以此为基础争取获得国家自然科学基金重点基金、杰出青年项目以及其他国家级项目资助;发表高质量(IF>10CellNatureScience系列) 的研究论文2-3篇,提高学术影响力。做好学科内和跨学科的合作,将基础研究成果与临床转化应用结合。

3.人才培养上:致力于临床医学、生物医学本科生与研究生的多方位培养,争取让本人指导的研究生再次获得“浙江大学优秀毕业生”等荣誉,积极帮助团队的其他教师继续获得国家级项目,培养一支学术作风严谨的高水平研究团队;

4.学科建设上:积极投入学科和系、院的公共建设,继续在医学院技术平台管理委员会和本人作为主要成员引进的李达三叶耀珍再生医学基金承担服务工作;

     5. 其他方面:积极参与国内外学术交流,在本人担任委员的中华医学会骨质疏松及骨矿盐分会基础组和国际华人骨研学会争取承担更大的责任,并在本人任职的浙江省生物医学学会(理事)和浙江省生化学会(副秘书长)做好服务工作; 继续做好学术杂志及基金的评审,以及科研普及与交流活动。此外,继续把本人作为主要创办者设立的“中国肌肉研究研讨会”办成国内最高水平的肌肉研究会议。
