二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

本人一直从事天然免疫和炎症应答的调控机制研究,前期的研究工作揭示了数个调控巨噬细胞抗病毒固有免疫和炎症因子产生的新机制,主要包括阐明了miRNA-92amiRNA-33E3泛素连接酶FBXW7对在巨噬细胞炎症反应和产生I型干扰素的重要调节功能及其分子机制。近5年以第一作者或共同第一作者在Nat Commun,J Immunol,J Biol Chem等SCI收录的主流期刊发表论文6篇,以参与作者身份在J Allergy Clin Immunol等SCI收录期刊发表论文10篇。作为负责人主持国家自然科学面上项目、国家自然科学青年基金、中国博士后科学基金、浙江省博士后择优资助各1项。

本人一直从事天然免疫和炎症应答的调控机制研究,并取得了一系列学术成绩。成果以第一/共同第一作者发表在Nat Commun,J Immunol,J Biol Chem等权威期刊。获得的主要创新性发现包括:

(1) 在模式识别受体触发的固有免疫和炎症应答的调节机制方面提出了新的学术观点,揭示了非编码RNA-MicroRNA和E3泛素连接酶FBXW7等分子调控炎症和干扰素产生的分子机制。本人的研究发现,巨噬细胞中TLR信号的激活可抑制miR-92a的表达,其可通过靶向抑制MKK4降低JNK/C-JUN信号通路的活化,最终抑制炎症因子的表达。该研究结果为巨噬细胞炎症应答的调控机制增添了新的认识,并可能为炎症引起的免疫相关性疾病的诊治提供新的靶标。本研究成果于2013年发表于J Biol Chem. 目前他引次数已达到46次,被Blood, J Clin Invest, Advanced Drug Delivery Reviews等期刊论文引用,入选ESI引用前3%论文。同时,我们的研究还发现巨噬细胞中TLR受体触发的I型干扰素可通过胆固醇25羟基氧化酶(CH25H)降低miRNA-33的表达。miR-33的下降释放了对靶基因胆固醇转运蛋白ABCA1 和 ABCG1 的抑制,从而促进细胞内游离胆固醇的外流,膜内脂筏含量降低,TLR信号减弱。该部分研究提示了胆固醇与炎症之间存在的关系,为炎症引起的免疫相关性疾病的诊治提供了新的思路。本研究成果于2016年发表于J Biol Chem。此外,我们利用E3泛素连接酶FBXW7选择性基因敲除小鼠研究发现,FBXW7在RNA病毒刺激后能从胞核转位至胞浆,结合并降解SHP2来抑制SHP2/C-cbl复合物对RNA病毒sensor—RIG-I分子的降解,从而促进I型干扰素的产生,促进抗病毒免疫应答。该研究揭示了FBXW7在抗病毒天然免疫中发挥着重要的调控功能,为深入认识天然免疫的调节机制提供了新的观点,并可能为治疗感染性疾病提供理论依据。本研究成果于2017年发表于Nature子刊Nature Communications.

(2) 揭示了肿瘤炎性微环境中重要的信号途径TGF-β/miR-494/AKT在调节免疫抑制性细胞(myeloid-derived suppressor cells, MDSCs)中的新机制。我们的工作发现肿瘤相关因子TGF-β可显著MDSC中microRNA-494的表达,miR-494可通过降低靶基因PTEN来调控Akt 信号途径,从而促进MDSC的聚集和抑制T淋巴细胞的功能,帮助肿瘤细胞实现免疫逃逸。该研究为阐明肿瘤的免疫逃逸机制提供了新的认识,而且miR-494有可能成为潜在的免疫治疗靶点。本研究成果于2012年发表于免疫学TOP期刊J Immunol,目前他引次数已达到101次,被Nature Reviews Cancer, Immunological Reviews述评及Immunity等期刊论文多次引用,入选ESI引用前3%论文。


三、岗位工作思路&= #21450;预期目标

聘期内工作思路:

作为学科建设的一位新成员,在聘期内将积极承担学院和药理学学科系的教学科研任务,包括课堂授课,学习教育教学理论和专业知识,参加各类教学教育实践活动,向经验丰富的老师学习,提高自己的教学水平,同时,充分利用网络信息资源,推进课程建设质量。

在原有的研究课题方向上继续深入,包括阐明IL-9在炎症性自身免疫性疾病如关节炎中的作用,以及miR-33、CLN3对巨噬细胞固有免疫应答的调控机制研究,尤其关注与免疫代谢的关系,力争产出创新性学术成果,并深入挖掘,发现和鉴定潜在的药物靶点,逐渐形成自己的研究特色。关注本学科国内外的最新进展,及时把握学科的发展方向和发展趋势,在已有的实验基础上,不断地探索和思考,逐渐积累研究经验,建立自己特有的前沿研究方向,达到国际先进和领先水平。同时,加强与国内外科研机构的交流合作。积极参加国内外学术会议,做会议报告,提高自己的能力以及积极听取其他专家的意见,促进科研项目的进展。作为负责人承担国家及省部级课题,争取产出具有一定影响力的创新性科研成果,为临床疾病的诊治提供新的思路和潜在靶点,力求将来实现成果转化。

积极参与学科建设规划的制定和实施,立足前沿,协助培养具有开拓创新能力的研究生,对学科建设和人才培养做出较大贡献。
