二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

 申请人任现职以来主要从事单细胞分析技术方面的研究。主要学术成就如下:(1)建立了一个低成本、高通量、完全国产化的全新的单细胞分析新技术,与公司合作,研发了一款试剂盒,初步实现产业化;(2)绘制了首个哺乳动物细胞图谱,该工作覆盖了小鼠8个不同生命阶段的主要的组织器官和细胞系,建立了囊括百余万个单细胞转录组的数据库,在此基础之上,建立了互动性的小鼠细胞图谱公共分析平台,可用于基因搜索和单细胞比对;(3)完成了首张人类细胞图谱,通过与小鼠细胞图谱的比较分析,发现并定义了多种哺乳动物的新细胞类型;(4)在细胞图谱数据的基础之上,从单细胞水平探索性解析了细胞分化过程中的细胞命运决定的机制。

本人自2017年12月起任职浙江大学医学院基础医学系副教授,一直致力于利用单细胞分析技术绘制哺乳动物细胞图谱、胚胎干细胞分化图谱、血液系统分化图谱等,专注于在单细胞水平解析干细胞、组织器官以及胚胎的细胞组成、细胞间协同作用、发育以及衰老等生命科学领域热点问题。目前主持国家自然科学基金优秀青年科学基金一项、面上项目一项、青年项目一项,参与国家重点研发计划干细胞与转化研究专项一项(个人经费200万);获国家发明专利2项;兼职中国老年学与老年医学学会抗衰老分会委员。任现职近2年来,以第一作者或通讯作者身份在CellGenome Biology等权威期刊发表学术论文共4篇,总影响因子64 ,单篇最高影响因子36.4。其主要学术成绩包括:

 (1)自主研发了Microwell-seq高通量单细胞测序平台,该平台具有成本低、通量高、全国产化、污染率低、不依赖昂贵仪器等优点,并利用这一平台绘制出了世界首个哺乳动物细胞图谱(小鼠细胞图谱)。该工作覆盖了50余种小鼠组织器官和细胞系,建立了大于40万个单细胞转录组的数据库。基于微孔板数据,建立了一个单细胞比对系统,可用于鉴定未知细胞的种类,检验体外分化、转分化、去分化细胞的质量,该系统兼容其他单细胞技术平台来源的数据。此外,我们还建立了小鼠细胞图谱公共分析平台,用于在线基因搜索和单细胞比对。这项工作于2018年2月以亮点论文的形式发表在顶级期刊Cell上,申请人为第一作者和共同通讯作者。该工作为单细胞组学领域里程碑式的研究进展,引起业内巨大,人民日报、新华社等媒体对其进行了重点报道,F1000推荐了该文章,评价为"very good",在发表后一年多的时间里,被CNS在内的顶级期刊引用百余次,被评为热点论文、高被引论文。

 (2)利用微孔板单细胞分析平台完成小鼠发育以及衰老各个阶段(胚胎9.5,胚胎13.5,新生1天,3周,6-8周,12个月,18个月,24个月)主要器官(脑、心脏、肾、小肠、肝、 肺、胰腺、胃、子宫和睾丸)的单细胞转录组图谱的绘制,建立了一个囊括90余万单细胞转录组的数据库,完善了小鼠图谱网络分析平台。基于图谱数据,建立小鼠发育和衰老的分子调控机制,解析了这一重要生命过程的细胞命运决定机制。目前该工作已经进入文章整理阶段,申请人是共同第一作者和共同通讯作者。

 (3)完成了首张人类细胞图谱,该工作覆盖了50余种正常成人和胎儿的组织和器官,建立了大于50万个单细胞转录组的数据库。通过与小鼠细胞图谱的比较分析,发现并定义了多种哺乳动物的新细胞类型。该研究覆盖了人体内各种主要细胞类型,是一个全面的数据化资源,惠及细胞生物学、发育生物学、神经生物学、系统生物学、临床医学等多个学科领域,将从单细胞水平推进细胞命运决定以及疾病发生机制的研究,并将对正常与疾病细胞的检测、分类和鉴定带来深远影响。目前该项工作已经进入了第四轮评审(Nature),申请人是第一作者和共同通讯作者。  

 (4)利用高通量微流控芯片C1 HT IFC在单细胞转录组水平解析人胚胎干细胞以及拟胚体三胚层分化的动态机制,构建了人胚胎干细胞早期分化图谱。候选人团队在人多能干细胞分化产生的拟胚体中发现了三胚层多种组织的前体细胞,包括神经、心肌、骨骼肌、内皮、肝脏、上皮等,通过时序性分析描绘了这些前体细胞分化过程中动态的基因变化。该研究将加深人们对早期胚胎谱系分化分子通路的了解,并促进分化方案的优化,有望解决目的细胞功能不完善等问题。这项工作于2018年4月发表在基因组学权威期刊Genome Biology上,申请人为第一作者。

 (5)从单细胞水平开创性的比较了物种间造血系统的差异,跨物种的单细胞比较组学分析更为系统的验证了传统寡能祖细胞的异质性,揭示了人鼠造血干细胞的分化机制。此外,我们还通过单细胞q-PCR、体外克隆形成能力、免疫缺陷小鼠异种嵌合能力等手段,高通量筛选出一个小分子化合物组合能够实现人造血干细胞体外的有效扩增,并通过单细胞测序技术解析这一过程的动态机制和相关信号通路。对人造血干细胞分化和体外扩增的研究,有望为人类血液系统的细胞治疗和基因治疗提供可靠的细胞来源。这两项工作分别于2018年7月和2018年10月在线发表在Cell Discovery上,申请人分别为最后通讯作者和共同通讯作者。

 综上所述,本人的学术成果归纳总结为:1)建立了微孔板单细胞技术平台,推动了单细胞测序进入低成本、高通量的时代;2)绘制了小鼠和人类等细胞图谱,建立了细胞图谱网络分析平台;3)探索性解析了细胞分化过程中的细胞命运决定机制。

1.教学方面:主讲不少于1门本科生课程,参与研究生教学,完成所在院系(单位)平均教学工作时数或基本教学工作量要求;积极参与本科生学术指导工作,例如国创、省创、SRTP(校、院级)、各类竞赛及本科生毕业设计等。

继续围绕单细胞分析技术方向开展工作,预期目标如下:

1)完成好已经获得的经费项目,包括国家重点研发计划、国家自然科学基金优秀青年基金、面上项目、青年项目等;

2)三年内以浙江大学作为第一单位,本人为通讯作者或第一作者发表3篇以上IF≥5的SCI论文或本领域Top期刊论文,或2篇以上IF≥10的SCI论文;

3)积极申报国家三大奖/省部一等奖;

4)积极申报国家和省部级各类基金;

5)申请国内授权发明专利1-2项,并推进成果产业化。

3.人才培养方面:

1)培养研究生3-5名;

2)指导、协助青年教师从事教学科研工作和申报省级和国家级科研项目,鼓励青年教师来本课题组参与部分科研项目。

4.学科建设方面:
