二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

本人作为PI带领多学科交叉科研团队长期致力于医学影像学研究,在磁共振新型对比剂的研发、肿瘤分子影像学及介入分子影像学等领域取得了丰硕科研成果。研发了2种磁共振新型对比剂,构建了10种恶性肿瘤靶向诊断、靶向治疗及诊疗一体化的纳米探针及诊疗方法,建立了影像引导的恶性肿瘤介入基因治疗方法,成功构建6株中国人群恶性肿瘤原代细胞系等。近5年以共同通讯作者或第一作者(包括共同)在国际著名期刊如Advanced MaterialsNature CommunicationsNano LettersBiomaterialsTheranostics等发表SCI论文17篇(4IF>10,最高IF25.8)。主持国家自然基金面上项目2项,主持省重点研发项目及省自然基金2项,作为骨干成员参与国家重点及重大项目3项,以第一发明人授权国家发明专利3项。

 本人长期从事影像医学的临床、科研及教学工作,拥有丰富的影像诊断及分子影像学研究经验。与临床学科、分子生物学、材料学、化学、药学等学科交叉与融合,推进新型影像对比剂的研发与恶性肿瘤靶向诊疗新技术研究。在新型影像对比剂的研发、恶性肿瘤的多功能分子影像靶向探针、靶向诊疗新技术及个性化治疗等方面取得相应的原创性科研成果,建立一支高素质的多学科交叉的分子影像学术团队,逐渐在该研究领域形成自己的特色。近5年以共同通讯作者或第一作者(包括共同)在国际著名期刊如Advanced MaterialsNature CommunicationsNano LettersBiomaterialsTheranosticsACS Macro LettersPolymer ChemistryNanomedicine等发表SCI论文17篇(4IF>10,最高IF25.8)。主持国家自然基金面上项目2项,主持省重点研发项目及省自然基金2项,作为骨干成员参与国家重点及重大项目3项,以第一发明人授权国家发明专利3项。主要学术成绩、贡献、创新点科学价值如下:

1.  新型MR对比剂的研发:

目前应用于临床的磁共振对比剂主要为钆离子(Gd3+)的配合物,但含钆对比剂的缺陷一是存在生物毒性,可引起肾源性系统性纤维化的风险外,且在脑组织内长期沉积;对比剂缺陷二是无肿瘤靶向性;缺陷三是肠道不吸收及无法通过肠道吸收增强成像。

本人带领团队设计合成了2种新型MR对比剂,(1)研发了一种含有聚多巴氨基酸螯合三价铁离子的两亲性聚合物纳米胶束(PDOPA-b-PSar),为新型可降解及可被人体利用、高效且无钆的MRI阳性对比剂,不仅有望替代传统Gd3+对比剂,而且可以同时负载化疗药及抑制剂,有望实现肿瘤靶向诊疗。研究成果以共同通讯作者发表于ACS Macro Lett,并申请了国家发明专利及PCT(申请号:201810468018.2)。(2)成功研制了肠道易吸收MR纳米对比剂,采用含Gd3+固体脂质纳米应用于早期结直肠癌,以肠道吸收的方式建立新型磁共振结直肠成像方法,填补目前影像学无法实现肠道吸收功能评价这一空缺,有助于炎性肠病及结直肠肿瘤的早期诊断,同时可为产业化研发集结直肠癌早期诊断、治疗及疗效评价于一体的新型纳米药物开发奠定基础。该成果以第一作者发表于J Biomed NanotechnolMagn Reson Med,并以第一发明人授权2项中国发明专利(ZL 201510573829.5ZL200910095903.1)。主要创新点与科学价值:两种新型MR对比剂的使用更安全,尤其第一种,无钆且可被人体降解利用;具有肿瘤靶向性,有助于肿瘤的鉴别诊断及靶向诊疗;建立新型磁共振结直肠成像方法,具有胃肠道靶向性成像及肠道吸收功能评价。

2.  肿瘤分子影像学研究:

肿瘤分子影像学研究包括肿瘤靶向诊断与诊疗一体化,其中诊疗一体化是指在同一载体上同时负载治疗药物和分子诊断探针,在诊断疾病的同时对疾病进行有效治疗的方法(肿瘤靶向诊断+靶向治疗)。因此,肿瘤分子影像学研究涉及肿瘤靶向诊断、靶向治疗与诊疗一体化。

1)肿瘤靶向诊断方面,设计合成螯合稀土的高分子材料,负载Gd3+MR成像)和Eu3+(铕,红色荧光成像)以合成纳米对比剂。该纳米对比剂具有较高的肿瘤靶向性,可以实现恶性肿瘤的双模态成像。研究成果以主通讯作者发表于Polym ChemIF:5.687,封面文章),双模态纳米对比剂制备与应用获中国发明专利ZL201510155268.7通过RAFT法可以简便合成结构可控的两嵌段聚合物PDKMA-b-POEGMA,该两亲性嵌段共聚物与稀土离子螯合后形成的纳米粒子生物相容性好,同时具有优越的荧光成像性能和MRI成像性能,在医药和生物领域,尤其对恶性肿瘤的诊疗具有较高的应用价值。

2)肿瘤靶向治疗方面,本团队基于3种纳米制备技术研发了5种恶性肿瘤靶向治疗纳米探针。单甘酯表面修饰的无定型碳酸钙纳米粒,负载模型药物盐酸阿霉素,该给药系统能够实现载体在酯酶丰富的肿瘤环境下特异性的快速释放药物,引发旁观者效应并促进药物向肿瘤深层组织递送,达到减少药物毒副作用,增强肿瘤治疗的效果,研究成果以共同通讯作者发表于Adv Mater(IF25.8)。本研究开发的给药系统通过肿瘤特异性的酯酶触发水敏感的药物释放,达到减少药物毒副作用,增强肿瘤治疗的效果,为高效、安全的肿瘤治疗制剂的后续开发提供了新的思路。

本团队研发了磷脂表面修饰的无定型碳酸钙纳米粒,负载模型药物米托蒽醌,实现了药物的有效负载和长期稳定,该给药系统能够实现叶酸受体靶向的肿瘤定位并实现肿瘤环境下特异性的快速释放药物,相比游离药物具有更强的肿瘤治疗效果。负载模型药物盐酸阿霉素,该给药系统能够实现肿瘤环境下水敏感式的快速释放药物,使负载的药物能够完全有效的发挥疗效,并促进药物向肿瘤深层组织递送,达到了减少药物毒副作用,增强肿瘤治疗的效果,研究成果以共同通讯作者发表于Chem Commun Int J Nanomed

此外,本团队还研发了壳聚糖硬脂酸聚合物负载中空金纳米及近红外荧光剂,表面修饰肿瘤靶向性多肽,实现恶性肿瘤靶向光热治疗。在此基础上开展了近红外光控制中空金纳米促进释放p53的肿瘤基因治疗,研究成果以共同第一作者及共同通讯作者分别发表于Biomaterials (IF10.273)Nanomedicine

3  肿瘤诊疗一体化方面,本团队研发了碳酸氧铋管状组装体介导肿瘤靶向成像并用于肿瘤放化疗协同治疗,该组装体可在微酸环境下解组装进而快速经肾清除,为高效、安全的肿瘤诊疗制剂提供新的思路,研究成果以共同第一作者发表于Nano Lett. (IF13.969);同时以新型可降解的多聚赖氨酸纳米负载Gd-DTPA和紫杉醇,开展了恶性肿瘤诊疗一体化的探索,实现了肿瘤靶向诊断及靶向治疗,研究成果以共同通讯作者发表于Sci Rep

 3.  肿瘤的介入分子成像方面:本团队首先完成了射频加热化疗提高乳腺癌疗效的研究:介入MR  分子影像引导下的乳腺癌化疗;然后采用射频加热结合热休克蛋白启动子介导的荷胆管癌瘤裸鼠HSV-TK自杀基因治疗,结果显示利用热休克蛋白启动子引导下的HSV-TK自杀基因治疗结合射频加热可以有效的抑制肿瘤增殖,并引发显著凋亡,为进一步开发胆道肿瘤的介入射频基因治疗体系奠定了坚实基础。研究成果以共同通讯作者发表于Theranostics (IF8.651)  Am J Cancer Res

4.  分子影像学相关的分子生物学研究:

1  中国人群恶性肿瘤原代细胞系构建方面,本团队经驯化培养获得永生化中国人群乳腺癌细胞系(ZJU-0327, ZJU-0725ZJU-1127)、胆囊癌细胞系(ZJU-0430)及胆管癌细胞系(ZJU-0826ZJU-1125)。经过种属、组织来源及恶性生物学特征的鉴定,建立的细胞系确认来源于相应组织,且不被目前存在的任何细胞系污染;同时分析了建立的细胞系的个性化生物学特征,对于相应疾病的发病机制和治疗研究奠定了基础。因此,所建立的肿瘤细胞系能够代表近年来中国乳腺癌、胆管癌和胆囊癌人群部分发病机制及生物学特征的相应癌细胞系。研究成果以共同通讯作者发表2SCI论文(Cancer Cell International)并以第一发明人授权国家发明专利(ZL201711264316.1)。

2)基础研究进展方面,首次揭示了Dbf 2相关激酶1(NDR1)以激酶活性非依赖形式促进体内外抗病毒天然免疫应答,研究成果以共同通讯作者发表于Nat Commun(IF13.811)  

本团队利用生物信息学手段,通过在线数据库和分子生物学技术发现Wnt5bBasal-like型乳腺癌中特异性高表达,且可通过激活经典和非经典Wnt通路以维持Basal-like型乳腺癌的表型。同时在实验动物身上,利用红色荧光与超声两种活体成像技术结合的方式证实WNT5B敲低或采用小分子LGK974抑制Wnt5b蛋白分泌可明显阻滞Basal-like型乳腺癌的生长。研究成果以共同通讯作者发表于Cell Commun Signal

5.  MR成像方法:3.0T磁共振上使用多翻转角T1 mapping评估肝纤维化,建立了肝纤维化MR评估方法,研究成果以共同通讯作者发表于J Magn Reson Imaging



三、岗位工作思路&= #21450;预期目标

岗位工作思路

1. 教学:积极参与本科生、研究生的授课,提高授课质量。积极引导与培养研究生、交叉学科研究生及博士后,加强下级医生的培训与教育,提升学生的临床与科研能力,为国家输送高层次复合型人才。

2. 科研:进一步推进多学科交叉合作,凝练与聚焦研究方向。发表我科主导的高影响因子及高引用SCI论文,申报国家及国际发明专利,积极促进临床转化;积极组织申报各类课题及项目,争取国家级项目连续资助,国家重点与重大项目取得突破;积极推进在研各类项目,确保项目顺利进行与高质量完成;协助科主任做好本科室各类项目申报与科研管理。

3. 临床:加强亚专科建设,积极推进影像新技术与新方法在临床中的应用,进一步提高影像诊断水平。积极参与院内外疑难病例会诊与讨论,与临床相关学科紧密联系与合作,积极参与多学科讨论与建设,提升MDT水平。

4. 学科建设:协助科主任做好科室学科建设、科学研究、实验室平台管理及人才培养。以双一流建设为引领,凝练科研方向及加强科研团队建设,积极引进及培养人才,加强人才梯队建设。增进及广泛开展国际交流与合作。

 

预期目标

1.  教学工作:认真完成学校课程教学与指导任务,开展相应的教育改革,所有的教学成绩达到优良。积极做好研究生培养工作,争取优博或优硕,博士生出国率达到100%。

2.  科研方面:年均申请国家级课题1-2项,力争3年内国家重点或重大项目取得突破,年均发表SCI论文5-6篇(1-2IF≥10),3年内申请省级或国家奖12项,年均申请国家发明专利或PCT 1-2项。

3.  临床方面:提升亚专科建设内涵,进一步细分亚专科,积极开展多学科联合读片,以提高影像诊断水平。积极推进影像新技术与新方法在临床中的应用,如人工智能影像组学辅助疾病诊断。

4.  学科建设:按照本学科发展规划实施,进一步加强实验室平台建设,促进医工交叉,在分子影像学及人工智能影像组学方面取得突破;培养优秀人才。加强国际交流,每年国际交流12次。
