二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

1. 成功构建了小鼠条件性社交恐惧模型,并进一步阐明了前额叶皮层内一条全新的去抑制神经微环路(SST-PV-pyramidal neurons)在社交恐惧行为中的调控作用,该研究成果以共同第一作者身份发表在 Neuron(IF: 16.3)

2. 发现慢性内脏痛导致星形胶质细胞病理性变化,星形胶质细胞-神经元L-型乳酸分子通路参与了调控慢性内脏痛相伴随的神经元功能变化及决策行为障碍,该研究成果以第一作者发表在Cell Reports(IF: 8.7)

3. 发现慢性内脏痛导致大鼠内侧丘脑-前扣带皮层(ACC)突触传递效能改变,并证明该通路长时程突触可塑性改变是慢性内脏痛在脑内的储存机制,该研究成果以第一作者发表在Cerebral Cortex(IF: 8.3)

4. 发现星形胶质细胞参与焦虑样行为表达过程中神经元功能的调控,作为项目负责人获得国家自然科学基金青年项目资助。

近年来申请人一直围绕情绪与认知功能的神经调控机制展开系列研究,特别是在病理状态下情绪及其相关的认知功能障碍的研究方面积累了一定的工作经验和理论知识。在神经科学前沿技术,特别是在体电生理技术,多通道电生理数据分析,光遗传等技术上受到了很好的训练。近5年来,已发表SCI收录的科研论文12篇,其中第一或共同第一作者论文7篇,第一作者论文主要发表在Neuron, Cell ReportsCerebral Cortex等神经科学高水平杂志。另外,申请人作为项目负责人主持国家自然科学基金青年项目1,并作为核心骨干成员参与了国家自然科学基金面上项目1项,国家自然科学基金/香港研究资助局联合项目1项,参与香港基础研究项目4项,香港医疗卫生研究基金1项,香港创新及科技基金1项。主要学术成果如下:

 

1. 社交恐惧行为的前额叶微环路机制:

社交恐惧主要表现为对正常社交行为或社交场合的焦虑和回避,是众多神经精神疾病的一个高度共有病征。例如抑郁症,自闭症等都伴随有社交恐惧症状,早期干预社交恐惧能减小引发其它精神疾病并发症的风险。此外,长期的社交恐惧会引发社交恐惧症,一种十分常见的精神疾病,严重影响着患者的日常生活和工作,给他们的身心健康造成了极大的危害,同时也给患者家庭和社会带来了沉重的经济负担。然而,其发生机制不甚清楚,且目前尚无令人满意的治疗方法。该研究中,我们自主研发了一套条件性社交恐惧造模系统。经历过社交恐惧条件化的小鼠对同类小鼠表现出强烈、持续的社交恐惧和社交回避反应。利用该小鼠模型,结合在体电生理记录、钙信号光纤记录、光遗传学、药理遗传学和动物行为学等一系列先进的实验方法,研究发现了前额叶皮层一条新的去抑制性神经环路(SST-PV-pyramidal neurons)介导小鼠的社交恐惧行为。该研究结果不仅揭示了前额叶皮层神经环路参与恐惧行为调控的机制,也为社交障碍的干预和治疗提供了理论指导。研究成果以本人为共同第一作者发表在国际神经科学顶级期刊NeuronIF: 16.3。文章发表以来受到了社会各界广泛关注。新华社、人民日报、中华人民共和国中央人民政府网、科技日报等众多媒体对我们的研究工作进行了报道。

 

2. 慢性痛相关的决策行为障碍

临床上慢性疼痛病人常伴有认知和精神方面的障碍,但其产生的机制仍不明确。我们建立了内脏痛大鼠模型,并研究了慢性疼痛相伴随的情绪与认知功能障碍的神经机制。研究中我采用大鼠赌博任务测试首先确定了慢性内脏痛大鼠有决策行为障碍,进一步的分子生物学机制研究发现星形胶质细胞来源的L-型乳酸参与了对慢性内脏痛伴随的决策障碍的调控。在ACC部位注射外源性L-型乳酸能治疗慢性痛大鼠决策行为障碍。为了确定星形胶质细胞-神经元代谢偶联中L-型乳酸这条通路是慢性疼痛伴随的决策行为障碍的产生机制,我们采用光遗传学手段特异性调控内源性星形胶质细胞,并发现在慢性内脏痛大鼠上光刺激激活星形胶质细胞可以释放L-型乳酸,并能基本恢复该病理动物决策行为障碍。这些研究结果表明星形胶质细胞L-型乳酸释放异常是慢性痛伴随的决策行为障碍的重要机制,星形胶质细胞-神经元之间L-型乳酸通路很可能是慢性痛伴随的决策行为障碍的治疗靶点。这些研究结果以本人为第一作者发表Cell ReportsIF: 8.7。国际脑研究组织(IBRO)主席、瑞士和欧洲科学院院士Pierre J. Magistretti教授在Nature Reviews Neuroscience杂志(Magistretti and Allaman, 2018),以及国际神经科学著名科学家、美国加州大学伯克利分校教授George BrooksCell Metabolism杂志(Brooks, 2018)分别发表综述论文对我们的研究工作做出了积极报道和正面评价。文章发表后受到香港成报、大公报、文汇报、星岛日报等媒体广泛报道。

 

3. 内脏痛脑神经机制的研究

慢性痛以什么样的神经信号储存在大脑的什么部位一直是痛觉研究领域未解决的难题。我们首先通过诱导大鼠肠道过敏反应建立了一种内脏高敏感动物模型以模拟临床上消化科常见的肠易激综合症。前期实验证明该动物模型在肠道炎症反应完全恢复以后仍然表现出对肠道扩张性刺激过度敏感,提示内脏痛由急性转化成慢性。采用在体诱发场电位分析突触传递及突触可塑性,我们发现内脏高敏感大鼠在疼痛神经处理通路上MT-ACC突触传递效率增高。同时,正常大鼠中MT-ACC可人工诱导出长时程增强(LTP),而在慢性内脏痛大鼠上诱导不出长时程增强。这些结果提示ACC突触传递增强是慢性内脏痛的关键机制。为进一步证明ACC突触可塑性与慢性内脏痛具有共同神经机制,我们在正常大鼠上使用高频刺激人工诱导LTP,发现这样的大鼠有内脏高敏感样的ACC神经元发放升高和对直结肠扩张刺激反应增强等现象,表明慢性内脏痛与ACC可塑性变化共享相同的神经处理机制。总之,我们的研究结果发现MT-ACC突触可塑性是慢性内脏痛信号储存的关键机制。该研究成果以本人为第一作者发表在神经科学领域著名杂志Cerebral CortexIF: 8.3。该论文发表后受到同行专家的较多关注,慢性疼痛领域多位专家在综述文章中引用我们的成果并给予正面评价。

 

此外,过去的研究表明疼痛导致ACC神经元发放速率升高,NMDA受体的NR2B亚基表达升高等生理病理性改变。然而,越来越多的研究表明神经信号的传递不仅仅依赖神经元发放速率的改变,网络同步性振荡、神经元动作电位与网络振荡之间的相位关系都编码着重要的神经信息。为了揭示疼痛与网络同步性振荡之间的关系,我们结合清醒动物多通道电极记录和神经电生理信号分析技术,对疼痛发生时网络振荡,特别是神经元发放与网络振荡相位锁定特性进行了系统的研究。我们发现反复直结肠扩张刺激诱导内脏疼痛发生时ACC场电位中theta波段能量增强,同时ACC神经元动作电位对其θ振荡发生相位锁定,并且MTACC之间的theta振荡同步性显著增强。这些实验结果表明,ACC网络振荡以及神经元发放与网络振荡锁相特性的改变是大脑中处理内脏痛信号的重要机制。该成果以本人为第一作者发表在神经科学领域老牌杂志Neuroscience

 

4. 病理状态下神经震荡异常的研究:

申请人在多通道神经数据分析方面接受了实验室长期合作伙伴香港城市大学电子工程系副教授陈皓敏博士(Dr. Rosa H.M. Chan)的训练。我们在多通道电生理记录与大规模神经网络数据分析,以及中枢神经参与动物痛觉、记忆及认知等多种行为学水平的机制方面取得了一系列研究进展,并有多篇论文通过同行评审发表在神经科学前沿杂志上。我们的研究表明前扣带回皮层及其与不同脑区之间theta振荡的同步性活动(synchronization)在化疗药物(Molecular Brain, 2015)、慢性雪卡中毒(Toxicology Research, 2017)以及母婴分离(Neurobiology of Learning and Memory, 2016)等多种动物疾病模型导致的记忆和认知功能障碍中起重要调控作用。


三、岗位工作思路&= #21450;预期目标

教学方面:

积极响应全国教育大会提出的“以本为本”的高等教育发展策略,坚持对本科教学工作的高度重视,积极参加生理系教学工作,包括工程生理学实验,生理科学等课程。根据自己在科学研究方面的经验积极引导学生设计生理学创新课题研究,扩展本科生知识面,培养学生学以致用的能力,将课本中的知识运用到一线科研工作中。同时,积极主动参与学校新教工始业教育培训,发现自身教学方面的不足并努力提升教学水平。积极向生理系教学指导教师和其他经验丰富的教师学习教学方法,并利用网络课程、多媒体教学等先进教学方法,努力积累教学经验和提升教学水平。根据自身教育背景,争取主讲或参与《生理学》,《神经生物学》等专业核心课程教学。争取每年指导本科生毕业设计课题1-2项,指导2-3名本科生参加创新性课题研究1项。及时高质量的完成学科分配的教学课时要求。此外,我将积极参与研究生科研指导工作,争取一年内开始培养硕士研究生。并将积极参与学科建设,人才梯队培养,主动参与学科规划和实验室管理工作。

 

科研方面:

青年教师是高校科研的生力军,进入副教授岗位后,我将一方面尽快完成现有课题研究工作,争取尽早发表高水平SCI论文1-2篇。另一方面,结合国际神经科学研究的前沿方向,自身研究背景和实验室研究基础,凝练新的研究方向,开展新的课题研究。近年来,社会行为的神经机制是国际研究的热点问题之一,解析社会行为的神经环路机制不仅填补了这一基础神经科学研究领域的空白,更将为临床上寻找社交相关精神疾病的干预手段提供理论指导。入职浙江大学以来,我们在社交恐惧行为神经机制方面的研究已经取得一定的突破和进展,研究成果发表在Neuron上,在此基础上未来的几年,我将利用小鼠作为研究对象,继续致力于研究社会交互行为的神经环路机制和社交障碍疾病的产生机理。在已展开的前期探索基础上,积极申请国家和省部级科研基金的资助,争取获得国家自然科学基金面上基金1-2项,并发表高水平SCI论文2篇以上。

此外,申请人前期的研究表明星形胶质细胞病变参与慢性疼痛相关的决策行为障碍,并阐述了其具体参与的分子机制:胶质细胞-神经元L-型乳酸分子通路,该研究成果发表在Cell Reports上。入职浙江大学以来,申请人结合前期研究积累,并依托所在研究组成熟的研究平台,采用钙测量,在体电生理,光遗传学,药理遗传学以及动物行为学等先进技术手段研究发现前额叶皮层星形胶质细胞在焦虑样行为表达过程中活动升高,并很可能通过对不同类型神经元调控作用的不同影响力周围神经网络活动并进而参与焦虑样行为的调控。该研究为胶质细胞参与行为调控提供新的思路,并为靶向胶质细胞治疗精神疾病提供了新的线索。该项研究也获得了国家自然科学基金的资助,申请人将在此基金的支持下,继续高质量完成项目研究工作,争取发表高水平SCI论文1-2篇。并计划以此为基础积极申请更多科研项目支持。

 

社会工作:
