 在眼科疾病相关�究中,申请人开�了遗传性白内障疾病相关基因��及其基因功能和晶状��育生物学基��究,在先天性白内障相关候�基因的突变方式及�病机制方��了大�的基��作。通�对晶状�遗传性疾病相关致病基因的克隆及其�白功能�究开�了一些开创性的�究�作,成功克隆了多个相关候�基因,�现白内障病因�究的新方向,在晶状��皮细胞自噬方�开创了新的�究方�并�行了首创性�究,已在�高影�因�的SCI  杂志,包括遗传学、眼科及细胞自噬等方�的高影�因�杂志Human Genetics, gene, plos one等杂志��表文章,为白内障的防�提�了相应的��依据。

利用候�基因直接���、连锁分析、外��组��、全基因组扫描和基因�白组学等前�技术�定出多个先天性白内障相关候�基因新突变和�年性白内障相关基因SNP位�,�表SCI�文阐�GJA8,GJA3,IGF1R基因与白内障的相关性并通�细胞实验等技术对基因�行功能�究,成为首先�现这些基因与不同�型白内障之间的相关性�并通�两项国�自然基�项目及多�SCI�文探究白内障相关致病分�机制,�缝隙连接�白50基因�手,�基因�除斑马鱼作为�式动物,�用细胞培养、免疫共�淀等��的实验技术,实验�现缝隙连接�白50基因对晶状��育�关�要,该基因�除后晶状�内自噬异常,同时首�探究了Cx50-Atg�白之间存在相关性�参与自噬的分�调控网�,并通�自噬�游mTOR通路抑制剂��实验探究其中的�物分��预��及��方案,�晶状�自噬角度探索白内障的�病机制,�找白内障新的�物防���,为临床�作提新思路。在无虹膜�系中�现多个新的PAX6基因突变位�,并通�细胞实验�究�实其内在的分�机制。�在�锥角膜疾病的人���中�现多个与之相关的新的基因突变位�,其中�现ZNF469基因与�锥角膜密切相关,首�在人�中�现91766559个新的ZNF469错义突变位�,并�行了动物�型及细胞实验验�探究其中的�能分�机制,为�锥角膜疾病的遗传学�究提����据。

   申请人作为��大学�学院附属眼科中心骨�成员之一,参与创新精准屈光性白内障手术技术,共同实现我国在世界白内障手术技术���的“领跑”地位�牵头制定白内障领域全�共�,规范了白内障防��程,建立11�市、县级基层技术示范基地及相关推广基地乡镇社区卫生院,��提升了基层眼科�疗���同时在白内障、�锥角膜及无虹膜等眼病中�现多个相关致病基因及�病机制,并全�系�揭示先天性及�年性白内障致病基因新突变及相关SNPs和�要�病机制,首��现白内障相关缝隙连接�白50基因参与自噬调控晶状�性状,�现其中的�物分��预��,为临床白内障的防�提�了切实�行的新思路。
