二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

应激与多种精神疾病密切相关。申请人通过连续性工作,明确了慢性应激引起促肾上腺皮质激素释放激素(corticotropin-releasing hormone,CRH)过度释放,持续激活其1型受体(CRHR1),导致海马、前额叶、旁嗅皮层等脑区突触发育和可塑性损害以及学习记忆障碍。进而,阐明了CRH-CRHR1信号通路的过度激活使突触细胞黏附分子nectin-1、nectin-3和钙结合蛋白calbindin表达下调、功能失衡,破坏突触可塑性,从而损害记忆。该系列工作突破了以糖皮质激素为主因的传统认识。此外,申请人亦发现:1)急性应激过度激活背侧海马CA1抑制性神经元并损害空间记忆;2)青春期NMDA受体慢性阻断可下调前额叶抑制性神经元parvalbumin和calbindin水平并损害逆转学习;(3)褪黑素可增强氟西汀的抗抑郁效果。上述结果为后续深入工作奠定了基础。

1. 率先发现应激开启CRH-CRHR1信号通路损害突触可塑性和认知的分子机制

在国外研究期间,申请人发现CRH-CRHR1信号系统在早年应激所致认知损害中发挥重要作用,初步阐明了CRH-CRHR1-nectin-3通路对突触可塑性和学习记忆的调节作用。在该研究基础上,申请人领导国内团队探索了CRH-CRHR1-nectin-3通路的下游分子,发现早年应激通过CRHR1与nectin-3调节海马钙结合蛋白calbindin的表达水平。更为重要的是,海马不同神经元亚群的calbindin分别参与调节应激易感性与抵抗性:兴奋性神经元calbindin表达降低介导了早年应激所致认知损害,而抑制性神经元calbindin表达降低则可对抗成年急性应激对认知的损害作用。此外,慢性应激通过激活CRH-CRHR1信号系统下调旁嗅皮层nectin-1表达水平并损害神经元形态可塑性。该系列研究发表于2017年10月Cell Reports杂志、2017年9月Translational Psychiatry杂志和2018年3月的Frontiers in Cellular Neuroscience杂志,被Annual Review of Genetics等杂志引用、评述,并被《健康报》等多家媒体报道,为应激相关精神疾病的机制解析与药物研发提供了新的思路。

 

2. 深入揭示早年应激通过CRH-CRHR1信号系统损害海马与前额叶发育

申请人领导国内团队从神经发育角度就早年应激损害神经元可塑性的机制开展了系列研究。结果表明,早年应激阻碍幼鼠海马CA3区锥体神经元树突与树突棘发育,损害海马环路组构与功能。持续拮抗CRHR1可以逆转早年应激对海马神经元发育的不良效应,而糖皮质激素受体拮抗剂则不具有治疗作用。以此为基础,研究团队明确了早年应激对不同年龄阶段的海马神经元结构发育的影响,发现早年应激通过CRHR1持续阻碍树突生长并增加青春期突触删除。这些结果更加充分地揭示了CRH-CRHR1信号系统是介导早年应激损害神经元发育的核心系统。

研究团队进而探索了早年应激损害小鼠前额叶锥体神经元发育的分子机制,发现早年应激可选择性阻碍背侧无颗粒扣带回皮层(ACd)第5层锥体神经元的顶树突发育与树突棘生成,且该效应可持续至成年期。相较而言,早年应激对于ACd第2/3层以及前边缘皮层锥体神经元的影响较为短暂,这表明早年应激对前额叶皮层发育的影响具有亚区和亚层特异性。药理学研究表明,CRHR1介导了早年应激对ACd第5层锥体神经元形态可塑性以及前额叶依赖性记忆的损害作用。

该系列研究揭示了小鼠海马与前额叶的形态发育关键期与应激敏感期,并首次从神经发育角度系统阐明了CRH-CRHR1信号系统是介导早年应激损害海马与前额叶发育和可塑性的核心应激因子。相关工作发表于NeuropsychopharmacologyBrain Structure and FunctionHippocampus杂志,目前已被NeuronMolecular Psychiatry等杂志引用、评述100余次。

 

3. 揭示海马CA1区抑制性神经元亚群参与应激效应的机制

研究团队发现急性应激可激活海马CA1区抑制性神经元,而采用化学遗传学手段选择性激活海马CA1区抑制性神经元可以模拟急性应激对记忆的损害。该成果于2018年4月发表在Neuropharmacology杂志。在此基础上,研究团队发现急性应激激活了海马CA1区CRH阳性抑制性神经元,该亚群神经元通过过量释放CRH,损害长期空间记忆。该部分工作尚在深入之中。


4. 其他工作

在开展应激研究工作的同时,申请人指导本科生与研究生探讨了抗精神病药和抗抑郁药的药理作用机制,从而探索精神分裂症与抑郁症行为障碍的细胞与分子机制,相关工作发表于Frontiers in Molecular NeuroscienceNeuroscience Bulletin等杂志。

 

综上所述,申请人通过系统性、连续性研究,阐明了CRH-CRHR1信号系统是介导慢性应激损害高级认知功能相关脑区发育与可塑性的核心因子,发现了突触细胞黏附分子nectin-1、nectin-3和钙结合蛋白calbindin作为CRHR1下游分子在应激障碍与学习记忆中的重要作用,提示了CRHR1拮抗剂具有治疗慢性应激相关认知障碍与精神疾病的潜在价值。该系列工作彼此联系、层层深入,推进了对应激相关精神疾病的分子神经生物学机制的认识,得到国际同行的认可与好评。

三、岗位工作思路&= #21450;预期目标

未来工作思路和预期目标如下:

1. 开展突触细胞黏附分子nectin-1参与应激所致焦虑障碍的环路机制研究,并以nectin-1为靶点开发模拟肽和阻断肽,走向药物靶点的临床转化。

2. 根据课题组前期发现的触觉丰富促认知、抗焦虑的结果(目前在Nature Communications杂志修回),深入揭示触觉及多模态感觉信息整合障碍参与应激相关精神疾病的环路和分子机制。

3. 结合深度学习和虚拟现实(如DeepLabCut、CAVE)等技术手段,打造国际一流行为学分析平台,并与相关课题组开展深入合作。
