二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

申请人入职以来,一方面致力于病人特异性iPSCs体外诱导再生晶状体研究,创立“荷包蛋”法诱导出世界上最大、透明、具有人晶状体类似结构及生物学特性、现今唯一具有光学特性的再生晶状体,并以此为基础在人晶状体发育、白内障病理机制、白内障新药研发等方面取得了一系列的成果及创新;另一方面致力于环境污染(特别是PM2.5)对眼睛疾病发病及病理机制研究,分别在基础研究及大数据分析两方面取得了一系列的成果及创新。任现职以来,依托浙大平台在上述两方面连续在Invest Ophth Vis Sci (IF=3D3.303, 眼科学TOP期刊)Environ Pollut (IF=3D5.009, TOP期刊),Mol Ther Nucleic Acids (IF=3D6.392),Medicine (IF=3D1.803  TOP期刊), Mediat Inflamm (IF=3D3.232)Endocrinology (IF=3D4.286, TOP期刊)发表第一作者论文5篇,共同第一作者论文2篇,主持国家自然科学青年基金1项,面上项目1项。

  1. 再生晶状体方向

白内障是全球首位致盲性眼病,现今唯一有效方法为白内障摘除联合人工晶体植入术,白内障治疗新药开发及再生晶状体治疗是未来白内障治疗的两大发展方向。几十年来,国内外学者尚未体外诱导出透明的、具有人晶状体类似生物学特征、规则结构及光学功能的再生晶状体。申请人入职以来一直致力于人iPSCs来源再生晶状体诱导新方法的探索,创立了“荷包蛋”法,诱导出世界最大、透明的、具有人晶状体类似生物学特性及结构、并且具有光学功能的再生晶状体(成果已发表在Invest Ophth Vis Sci, IF=3D3.303,眼科学TOP期刊;部分结果被2015ARVO年会选为大会发言,摘要收录于Invest Ophth Vis Sci, IF=3D3.303,眼科学TOP期刊。同时,以此为基础,a. 探索lncRNA调控自噬在人晶状体发育中的分子机制(成果已发表在Mol Ther Nucleic ACIDS(IF=3D6.392),官网online;部分结果被2017ARVO年会选为大会发言,摘要尚未收录);b. 构建病人特异性再生晶状体即病人特异性白内障疾病模型,探索特定突变诱发白内障病理机制,结合CRISPR-Cas9技术探寻可能治疗手段(部分成果由申请人协助指导的博士研究生投稿2016ARVO年会,并选为大会发言,摘要收录于Invest Ophth Vis Sci, IF=3D3.303,眼科学TOP期刊)c. 以上述病人特异性白内障模型为基础,构建稳定白内障新药筛选平台,并已初步开展相关药物筛选(成果尚未发表,为未来主要研究方向之一)。现将主要研究成果和具体创新点阐述如下:

1)再生晶状体诱导新方法---“荷包蛋”法的创建

晶状体再生一直是晶状体研究领域的重点和难点。虽然长期以来国内外学者不断提出“ SIDA”法、三步法等晶状体诱导新方法,但无论哪种方法诱导而成的再生晶状体都不理想,仅表现部分生物学特征,在结构及功能上跟体内晶状体相差甚远。申请人在前人研究的基础上,利用iPSCs诱导技术,结合晶状体发育已知分子通路,通过在特定时间点对晶状体发育祖细胞的定向分离,最终在体外将个体特异性iPSCs诱导成与人晶状体类似的、直径3mm左右(成人正常大小为直径9mm)的透明再生晶状体即晶状体小体(lentoid bodies LBs)。电镜、H&E染色、亚甲蓝染色、DiOC6染色表明成熟LBs具有人晶状体类似的囊膜、排列规则的晶状体上皮细胞、分化中的纤维细胞及成熟的纤维细胞;免疫荧光染色表明,不仅成熟LBs表达晶状体特异性标志晶状体蛋白abγMIP等,在LB分化过程中还表达placodal标志物SIX1EYA1DLX3PAX6,早期晶状体发育标志物SOX1PROX1FOXE3、晶状体蛋白aAaB。更重要的是,我们发现成熟LBs具有1.7倍左右的光学放大功能,为世界首创。由于LB分化过程中 “荷包蛋”样细胞结构的出现及对LB最终形成的关键作用,我们将该方法命名为“荷包蛋”法。该研究结果2017年以第一作者发表于Invest Ophth Vis SciIF=3D3.303,眼科学TOP期刊),部分结果被2015ARVO年会(美国丹佛)选为大会发言,申请人被授予当年ARVO年会的Travel Grant,得到国内外同行专家的高度评价。该成果被“钱江晚报”、sina新闻等国内多家权威媒体报道(题为“尿液里提取干细胞,有助于白内障诊治”),受到国内外广泛关注。“荷包蛋”法也已申请专利,待批。


2lncRNA调控自噬在人晶状体发育中的分子机制研究

由于伦理限制、人源性模型的缺乏,人晶状体发育分子机制一直都不清楚,而lncRNA在晶状体发育、白内障发生发展中的作用国内外仅有少量报道,几近空白。申请人利用“荷包蛋”法LB诱导过程作为人晶状体发育模型,首先通过跟小鼠发育中晶状体自噬过程的比较,证实“荷包蛋”法诱导过程能很好的再现晶状体发育过程;其次,通过micoarray技术及相关生物统计学分析发现,成熟的LB具有与在体晶状体类似的mRNA表达谱;随后,通过microarray技术还发现了对人晶状体发育具有重要作用的lncRNA,部分在人胚胎期晶状体中得到证实;最后,申请人通过lncRNA定位、knockdown等技术还发现lncRNA ALB能通过调控LC3B的转化来影响自噬从而实现晶状体发育中细胞器细胞核的退化。该成果为世界上首次对lncRNA在晶状体发育中功能及机制相关研究,以第一作者发表于Mol Ther Nucleic ACIDSIF=3D6.392),官网online,同时部分结果被2017ARVO年会(美国巴尔的摩)选为大会发言,受到国内外专家广泛的认同。


3)病人特异性白内障模型的构建及CRISPR-Cas9技术的运用

   先天性白内障很大一部分由基因突变引起,其治疗长久以来都是白内障领域的难点,基因修复及晶状体再生为未来主要发展方向,而CRISPR-Cas9技术的出现为其提供了可能性。利用申请人所在浙二眼科中心丰富的临床病例及前期在先天性白内障新基因新突变研究工作中的积累,申请人首先构建了先天性白内障病人特异性iPSCs,并以“荷包蛋”法为依托,在体外诱导成具有与先天性白内障病人类似的混浊的LBs,成功构建了病人特异性白内障体外模型。该成果由申请人指导博士研究生(协助指导)秦祯蔚投稿2016ARVO年会(美国西雅图),并被选为大会发言,会上受到领域内权威专家的认可。此结果争取年底投稿。在上述结果的基础上,利用CRISPR-Cas9技术,试图修复相关突变基因,并将修复好iPSCs诱导成LBs,为先天性白内障的治疗提供理论依据。现已基本建立CRISPR-Cas9技术平台,有望明年完成基因修复并投稿。


4)建立稳定白内障新药筛选平台

蛋白聚集导致晶状体混浊是白内障形成的基本病理基础。近期国内外研究者发现羊毛甾醇及其衍生物具有逆转蛋白聚集从而治疗白内障的作用,但由于人源性白内障疾病模型的缺乏,而一直停留在细胞模型、动物模型的研究上。利用荷包蛋法诱导而成病人特异性白内障模型为羊毛甾醇及其衍生物疗效的研究提供了完美的平台。申请人团队已经筛选出与羊毛甾醇结构类似的天然小分子,同时人工合成类似小分子,并通过在病人特异性LB上的作用,发现有部分新药能部分逆转病人LB的混浊程度,提示具有治疗白内障的潜能。该部分内容已获得初步成果,未来将结合动物模型,为进一步临床实验提供理论基础。


2. 环境污染(PM2.5)对眼睛疾病发病及病理机制研究方向

    环境污染特别是PM2.5对人体健康的影响越来越受到政府及社会的广泛关注,明确两者的关系、寻找特异性治疗手段成为关系到每个人切身健康的大事。眼睛作为人体除呼吸系统、皮肤外直接暴露于空气的器官,空气污染物很有可能影响眼睛疾病特别是眼表疾病发病及病程,但其相关性及病理机制都不清楚。申请人入职以来致力于以下两方面研究:a. 门诊量与个体暴露大数据分析,其中结膜炎和环境污染物相关性研究成果2017年以第一作者发表于Environ Pollut (IF=3D5.009, TOP期刊),干眼和环境污染相关性研究成果积极投稿中; b.PM2.5诱发角膜炎病理机制研究,其中PM2.5诱发自噬参与角膜炎发病的研究结果2017年以第一作者发表于Environ Pollut (IF=3D5.009, TOP期刊) 现将主要研究成果和具体创新点阐述如下:

1)眼表疾病就诊量和空气污染物浓度之间的相关性研究

依托浙二眼科中心庞大的门诊量,结合合作单位浙江省疾病预防控制中心、浙江省环保中心气候数据、空气污染物数据采集系统,从生物统计学、大数据分析的角度研究眼表疾病如结膜炎、干眼等就诊量和空气污染物的关系。研究发现,PM10, PM2.5, SO2, NO2, CO浓度的增加都将引起结膜炎就诊率的增高。该成果2017年以第一作者发表于Environ Pollut (IF=3D5.009, TOP期刊)(接收,pubmed online,web of science尚未收录)。研究还发现,PM10, PM2.5, SO2, NO2, CO浓度的增高跟干眼的就诊率也成正相关。该成果正积极投稿中。


2PM2.5诱发角膜炎病理机制研究

    虽然我们的研究及其他相关研究表明PM2.5能诱发眼表疾病,但其病理机制至今尚不清楚,国内外仅有少量相关报道。申请人研究首次发现PM2.5能通过调控自噬过程影响人角膜上皮细胞损伤,且存在时间和浓度依赖性。我们发现早期自噬作为细胞保护程序被PM2.5抑制,因此早期激活自噬可作为角膜炎的潜在治疗靶点;晚期自噬作为细胞损伤程序被PM2.5大大激活导致细胞损伤直至死亡。该成果2017年以第一作者发表于Environ Pollut (IF=3D5.009, TOP期刊)


 

 

三、岗位工作思路&= #21450;预期目标

教学方面:继续积极协助PI指导硕士生和博士生完成课题研究,争取明年获得硕士研究生招生资格,预期目标每年招收硕士研究生1名。

 

科研方面:一方面以“荷包蛋”法诱导病人特异性再生晶状体为平台,继续深入研究人晶状体发育机制、白内障病理机制,争取研发1-2种白内障治疗新药;另一方面继续在大数据分析及基础研究两个方面深入研究环境污染(PM2.5)跟眼部疾病的关系及分子机制,探索环境污染特异性眼睛疾病治疗新靶点。争取3-5年内在本领域高水平杂志上发表论文2-3篇,再次获得国家级和省级科研项目资助2-3项,申请发明专利1-2项,每年参加相关领域国内及国际会议1-2次,加强国际交流及合作。

 

重点实验室建设:继续协助浙江省重要致盲眼病防治技术研究重点实验室主任做好实验室日常建设、年度总结、创新平台对外服务等相关事宜。
