二、主要学术成就<= /span>

2.1 <= /span>标志性成果(不&#= 36229;过300字)

申请人一直从事干细胞与再生医学领域的研究,主要集中在运用干细胞与组织工程技术对中枢神经损伤(脊髓损伤)进行治疗修复及其机制研究。主要研究创新:1 分离培养获得眼眶来源的脂肪干细胞与17-beta-雌二醇联合运用修复脊髓损伤。用细胞转分化技术获得新的种子细胞来源,对其转分化机制利用RNA-seq和ATAC-seq等新技术进行解析。2 利用药物缓释给药系统投递药物及化学小分子等改善损伤病理微环境促进SCI修复,并对其病理微环境进行解析。发表SCI论文8篇,其中第一作者和通讯作者各一篇。作为负责人主持国家自然科学基金青年基金、浙江省创新团队子项目、紫金计划项目各一项;参与多项“863”、国家自然科学基金面上项目、国际合作项目。多次在国内外学术会议做报告或墙报展示,受到与会专家的认可和好评。

近年来申请人研究一直从事干细胞与再生医学领域研究,主要集中在运用干细胞与组织工程技术对中枢神经损伤(脊髓损伤)的进行治疗修复。一方面寻找合适的适用于中枢神经损伤修复的种子细胞,另一方面通过材料、药物、化学小分子等干预手段改善损伤病理微环境,提高损伤局部细胞存活、分化、迁移能力,提高干细胞治疗的效率,以期二者协同修复脊髓损伤,探讨治疗中枢神经系统损伤的新策略与方法。在基础医学领域的关键学会期刊发表SCI 多篇,其中第一作者和通讯作者各一篇,目前主持国家自然科学青年基金一项,浙江省科技厅重点科技创新团队子课题一项,作为主要骨干参与多项研究相关国家自然科学基金面上及重点项目和科技部的国际合作项目的研究,已主持并完成浙江大学优秀青年教师资助计划紫金计划。参与撰写、翻译两部(《再生医学基础与临床》与《神经病学彩色图谱》)教材。现将主要研究成果和具体创新点阐述如下:

脊髓损伤(SCI)是人类所经历的创伤中最具破坏性的创伤之一,常常导致永久性残疾,多由车祸、坠落等高能量撞击引发脊柱骨折、脱位所致。脊髓损伤预防保健防治基金会统计,目前在世界范围内,大约250万人受此病影响,每年有超过13万的新发外伤性脊髓损伤病例。其中只有不到1%的患者能够完全恢复,绝大多数患者均遗留部分或完全性瘫痪,损伤后患者的生活质量严重下降。而且在这些患者中82%是青壮年男性,给社会和家庭造成了不可估量的损失。由于SCI缺乏有效的治疗方法,由此带来的功能丧失往往伴随着病人的一生。

1 寻找合适的用于SCI修复的种子细胞

1)干细胞与组织工程的发展是目前SCI非常有前途的治疗策略。在成年哺乳动物中,虽然存在神经干细胞,但其对脊髓损伤的修复能力非常有限,因此仅靠机体自身的能力无法修复脊髓功能。没有合适的种子细胞以及移植后细胞的存活率低,在目前的SCI治疗中仍然是一个重大的挑战。因此寻找合适的种子细胞以及创造合适的环境促种子细胞存活对于脊髓损伤的治疗是非常重要的。之前的研究报告表明,从神经嵴的衍生组织中所分离出的眼眶来源的脂肪干细胞hEASCs能自发表达许多神经细胞相关的基因和蛋白,且具有多系分化潜能。目前研究中尚未有人使用hEASCs治疗脊髓损伤。另有研究表明脊髓损伤后早期投入化学药物17-beta-雌二醇治疗可以有效的阻止二次损伤的蔓延,最大程度的保护损伤后剩余的神经组织和控制炎症,减少空洞面积和减少细胞凋亡。因此我们提出假说,联合给药17-beta-雌二醇与hEASCs移植,是否会有利于脊髓损伤修复,促进大鼠瘫痪模型中功能恢复,相关结果已发表在top期刊Journal of Cellular and Molecular Medicine(影响因子=3D4.398)。

2)最近研究发现通过化学小分子可使成纤维细胞进行程序性编程转分化为目的细胞,根据文献的报导及我们实验的筛选摸索,发现了四种小分子可使小鼠成纤维细胞转分化为类神经干细胞样细胞,目前正在进行这部分的实验工作,期望获得更为方便及转化效率高的神经干细胞用于SCI 的修复治疗,并利用新技术RNA-seq和ATAC-seq测序深入解析其转分化的分子机制和重要信号通路,目前对此课题的研究较有信心,期望研究结果发表在高影响因子的杂志上。

2 改善SCI病理损伤微环境,提高局部原位细胞以及移植细胞的存活率

1)受损脊髓病理改变包括两个过程:原发性损伤和继发性损伤,主要造成不同程度的细胞死亡、组织水肿、胶质疤痕增生和脊髓运动功能丧失。而死亡的神经元裂解释放的毒素,损伤部位的慢性脱髓鞘等病理改变又可导致损伤平面上下两侧的脊髓组织坏死,导致损伤平面以下感觉和运动功能障碍。病变坏死区形成的空洞、反应性胶质疤痕增生、以及轴突断裂和脱髓鞘反应,也对神经细胞和轴突的再生产生抑制作用,新生的轴突不能通过瘢痕组织。因此胶质疤痕的增生是阻碍脊髓损伤修复的重要阻碍。因此我们设计纳米微球缓释系统进行局部药物缓释,改善局部炎症环境。flavopiridol是一种细胞周期抑制剂,在中枢神经损伤后可减少神经元和少突胶质细胞死亡,有效降低星形胶质疤痕形成。在动物实验中提示给药flavopiridol可明显提高运动功能的恢复。我们采用纳米微球损伤局部缓释flavopiridol,通过抑制星形胶质细胞增殖,减少胶质疤痕,促进脊髓损伤恢复,相关结果已发表在Biomaterials(影响因子=3D8.946)。

2)另外针对损伤局部胶质疤痕增生和大量炎症反应致细胞移植后存活率低、迁移能力弱的问题,我们体外实验发现FTY720能显著促进神经干细胞迁移和存活,抑制星形胶质细胞迁移。且有研究报导全身给药FTY720可促进SCI修复,但机制不清且全身给药有副反应。因此我们采用聚合材料PCL局部缓释FTY720复合多能干细胞来源的神经干细胞(iPS-dNSCs)移植能有效促进SCI修复,相关实验结果相关结果作为唯一通讯作者已发表在Biomaterials(影响因子=3D8.946)。

目前正对SCI病理微环境进行单细胞数据和蛋白芯片解析,找出SCI关键促炎因子。

  综上,申请人人任现职以来一直围绕脊髓损伤的干细胞组织工程治疗策略,对其可能的种子细胞及改善病理环境进行深入研究,为临床的应用提供前期理论研究基础;申请人目前已经取得了一系列阶段性科研成果,已经具备了独立开展科研工作的能力。申请人目前还有一些课题进展良好,预计近期将会不断取得新的科研成果,有信心在未来几年做出更扎实出色的成绩。

 


三、岗位工作思路&= #21450;预期目标

1. 教学方面:继续承担并主讲本科生或研究生课程的教学任务;保质保量地完成教学工作,积极参与青年教师教学技能的培训及竞赛,积极学习参与国际校区IBMS课程的讲授。每年指导硕士生一名,协助指导本课题组研究生课题及本科生毕业设计、SRTP等项目研究。争做一名传道授业解惑的优秀教师。


2. 科研方面:专注攻克本领域的关键科学问题:一方面,借助RNA-seq基因分析技术,ATAC-seq解析特定干细胞体外培养中表型丢失的分子机理及表型维持。另一方面,建立高通量平台筛选适合特定干细胞表型维持及分化的物理微环境刺激和化学小分子。争取3-5年发表高质量论文1-2篇。再次获得国家级和省级科研项目资助1-2项。
